木瓜樣蛋白酶是半胱氨酸蛋白酶家族,代表溶酶體蛋白水解系統的主要成分。通常,組織蛋白酶包含一個信號肽,然後是一個前肽,再然後是一個催化活性成熟區。組織蛋白酶F前體非常長的(251個氨基酸殘基)前區包含一個與組織蛋白酶L樣酶的前段相似的C端結構域、一個50個殘基的柔性連接肽和N端結構域預計採用胱抑素樣摺疊組織蛋白酶F前區在木瓜蛋白酶家族半胱氨酸蛋白酶中是獨一無二的,因為它包含這個額外的N端片段,預計與胱抑素超家族的半胱氨酸蛋白酶抑制劑具有結構相似性。這種胱抑素樣結構域包含一些已知對抑制活性很重要的元素。CTSF編碼預測的484個氨基酸的蛋白質,其中包含19個殘基信號肽。組織蛋白酶F含有五個潛在的N-連接糖基化位點,它可能通過甘露糖6-磷酸受體途徑靶向內體/溶酶體區室。組織蛋白酶F基因普遍表達,它定位於染色體11q13,接近編碼組織蛋白酶W的基因。[8]
泰國肝吸蟲的免疫診斷
組織蛋白酶F可用作測試泰國肝吸蟲造成的疾病的替代方法。[9]近年來,泰國肝吸蟲的診斷一直通過大便檢查來完成,被認為是「金標準」方法。然而,基於糞便的診斷可能是不可靠的[10],儘管它是一種非侵入性方法。進行的實驗表明,通過使用重組DNA技術和組織蛋白酶F半胱氨酸蛋白酶,可以產生更可靠的結果。
組織蛋白酶F在蝦夷盤扇貝中的表達
最近的實驗表明在蝦夷盤扇貝中發現了組織蛋白酶F。[11]在蝦夷盤扇貝的各個胚胎發育階段鑑定組織蛋白酶F基因具有重要意義。組織蛋白酶F基因在先天免疫反應中具有重要作用。蝦夷盤扇貝一直因細菌性疾病而遭受高死亡率。組織蛋白酶F的發現可以領導進一步的研究和解決蝦夷盤扇貝死亡率問題的方法。
